For highly lipophilic drug candidates or other test substances that tend to bind very strongly to plasma proteins it is difficult to assess the degree of protein binding and to rank compounds during lead optimization. Standard protein binding assays cannot be used to discern free fractions below the 1%-level. However, our high protein binding assay is able to discern differences in protein binding in the order of 0.01%. This assay is a variant of the erythrocyte partitioning assay using full plasma and TRANSIL beads.
